In Vivo Electroporation mediated DNA Immunization

Electroporation also refers to electropermeabilization or EP for short. Through the appropriate electrical stimulation of target cells, this technology can induce the transient transfection and increase cellular uptake of interested DNA. It has been widely used for in vitro transfection (e.g. bacteria, yeast, and plant protoplasts transformation) but can also be a qualified in vivo DNA delivery method. The in vivo EP starts with the administration of interested DNA by intramuscular or intradermal injection and then immediately applies electric pulses to the injection site by using appropriate device.

For genetic immunization, in vivo electroporation is a reliable approach which can strongly improve the delivery and duration of interested DNA into the cell. During the directly DNA injection of host animals, a lower immune response may be suffered due to the insufficient DNA transfection efficiency. Electroporation, in this case, can be a potential solution to overcome the limitation. This technique can transiently enhance the permeability of the cell membrane to increase the transfection efficiency. During the delivery, this approach can also lead to mild inflammation which contributes to the influx of antigen-presenting cells (APCs) and the exposure of the expressed foreign protein. On account of fewer resident APCs exist in the muscle tissue (compared with the skin), the induced inflammation via in vivo electroporation likely achieves much higher transfection efficiency and more robust immune response in comparison to the direct intramuscular/intradermal injection, especially for the immunization of large animals. In fact, in vivo electroporation can increase the expression of exogenous gene encoding protein 10 to 100 folds more than a direct injection of naked DNA.
